摘    要:探讨大块异种骨的制备及其在修复节段性骨缺损中的作用。方法将大块的牛松质骨制备成消除抗原性的载体,与牛骨形态发性蛋白(BMP)组合构成去抗原牛松质骨块/bBMP复合材料,采用兔桡骨15mm节段性骨缺损模型,研究去抗原质骨块/bBMP复合材料在修复骨缺损中的作用。

Abstract:AIM The purpose of this study was to develop a kind of xenogeneic bone graft material which could be used in the treatment of segmental bone defect. METHODS The antigen extracted big bovine cancellous bone blocks were harvested and impregnated with bBMP. A 15 mm defect non union model was established in the mid part of the radial diaphysis in rabbits and was used to study the effect of the composite grafts on the healing of the segment defect. RESULTS Abundant chondral and new bone islands were found in the defects 4 weeks after the operation and new bone formation was seen by the 8th week. Remodeling of new bone and resorption of the grafts implanted were observed by the 12th week. An early medullary canal containing marrow like substances formed by the 16th week in all the 8 defects treated with the composite grafts. Histomorphomentrical and radiographical evidence of complete healing was detected by the 20th week postoperatively, with the establishment of normal diaphysial structure and no remnant of the graft. CONCLUSION The antigen extracted bovine cancellous bone block/bBMP composite graft has the capability of healing large segmental diaphysial defects through both osteoinductive and osteoconductive mechanism.
