莪术二酮对大鼠血瘀模型血液流变学指标的影响

摘    要:目的观察莪术二酮(curdione)对大鼠血瘀模型血液流变学与纤溶指标的影响,同时研究其体外溶栓作用,从而探讨其活血化瘀功效的作用机制。方法以肾上腺素与冰水刺激为致瘀因素复制大鼠血瘀证动物模型,观察莪术二酮对血瘀证模型全血黏度及血浆黏度变化,检测纤维蛋白降解产物(FDP)和D-二聚体(DD);体外测定对不同时间大鼠血栓的溶解百分率,观察莪术二酮溶栓作用。结果莪术二酮可减低血瘀模型大鼠全血黏度和血浆黏度,改善血液循环(P<0.05);能降低DD水平;但对FDP水平无影响(P>0.05),且对体外血栓的溶栓作用不显著。结论莪术药材的"行气破血"作用与莪术二酮改善血液流变学指标有关,但实验剂量下莪术二酮溶栓作用不显著。

Effect of curdione on hemorheological indexs in rats with blood stasis syndrome

Abstract:Objective To study the influence of curdione on the hemorheological changes in blood stasis model rats and thrombolysis in vitro. Methods The model of blood stasis rats were induced by putting the rats into ice-water between epinephrine hydrochlaride twice of 0. 08 ml/100 g. With the models, the effect of curdione on hemorheology such as whole blood viscosity, blood reduction viscosity and plasma viscosity were detected. And PDF and DD were measured. In vitro, thrombolytic effects of curdione were determined by measuring the percentage of thrombolysis at different time. Results Curdione could significantly reduce the hemorheological indexes in different degrees, such as whole blood viscosity and plasma viscosity in blood stasis rats (P 〈 0.05). Curdione could change DD level, but it could not change FDP level in blood stasis rats ( P 〉 0.05 ). In vitro, curdione only possessed thrombolytlc effect in dose of 0. 235 g . L-1 and 2.35 g . L-1 , however, the low dose of curdione had not thrombolytic activity. Conclusion Curdione can obviously promote blood circulation to remove blood stasis. But curdione has not the notable activity of thrombolysis.
