胃癌组织中Her-2/neu、MMP-2和Leptin的表达及临床意义

目的探讨Her-2/neu、基质金属蛋白酶-2(MMP-2)和瘦素(Leptin)在胃癌组织中的表达及临床意义。方法分别采用免疫组化(SP)法检测80例手术切除的胃癌组织及癌旁组织中Her-2/neu、MMP-2和Leptin的表达水平。结果胃癌组织中Her-2/neu、MMP-2和Leptin阳性表达率分别为30%、62.5%和70.0%。Her-2/neu在癌旁组织中无表达,MMP-2和Leptin在癌旁组织中阳性表达率分别为37.5%、35%;两组差异有统计学意义(P0.05)。Her-2/neu、MMP-2和Leptin的阳性表达与浸润深度、淋巴结转移和TNM分期显著相关(P0.05),与患者的性别、年龄、肿瘤大小、肿瘤部位和肿瘤分化程度无统计学意义(P0.05)。三项指标之间的表达呈正相关(r=0.338,r=0.429,r=0.451,P0.05)。结论 Her-2/neu、MMP-2和Leptin在胃癌组织中的高表达与胃癌的发生、发展及转移密切相关,对指导胃癌靶向治疗有意义。

Expression and clinical significance of Her-2/neu,MMP-2 and Leptin in gastric carcinoma

Objective To investigate the expression and clinical significance of Her-2/neu,MMP-2 and Leptin in gastric carcinoma tissue.Methods The expressions of Her-2/neu,MMP-2 and Leptin were tested by immunohistochemical technique in 80 cases of gastric cancer and adjacent normal tissue.Results The positive rates of Her-2/neu,MMP-2 and Leptin in gastric carcinoma were respectively 30%,62.5% and 70%.No Her-2/neu was expressed in adjacent normal tissue.The positive rates of MMP-2 and Leptin in adjacent normal tissue were respectively 37.5% and 35%,and there was significant difference between two groups（P0.05）.The positive expression rates of Her-2/neu,MMP-2 and Leptin were correlated with the depth of invasion,clinical stage,lymph node metastasis（P0.05）.Their expression was not correlated with sex,age,size,site of tumor and the degree of differentiation（P0.05）.A positive correlation was found between expression of Her-2/neu,MMP-2 and Leptin（P0.05）.Conclusion High expressions of Her-2/neu,MMP-2 and Leptin were significantly correlated with development,invasion and metastasis of gastric cancer.Combining detection of Her-2/neu,MMP-2 and Leptin expression is meaningful to guide molecule targeted therapy.
