链脲菌素制备2型糖尿病大鼠模型的研究

摘    要:目的优化建立操作简便、条件稳定、成模率高、造模周期短、具有胰岛素抵抗的2型糖尿病模型。方法 6周龄大鼠以高糖高脂饲料喂养4周后,禁水禁食18 h,一次性腹腔注射1%链脲菌素(STZ)溶液35mg·kg-1,注射后继续禁水禁食1 h;对照组腹腔注射相同剂量柠檬酸-柠檬酸钠缓冲液。结果 STZ注射后14 d空腹血糖值稳定,口服葡萄糖耐量实验符合糖尿病标准,空腹胰岛素水平接近对照组,成模率93%,模型死亡率0%。结论成功制备了2型糖尿病模型,优化了2型糖尿病造模过程及提高了成模率。

Studying of streptozotocin inducing type 2 diabetes rat model

Abstract:Objective To establish a simple, stable, quick, type 2 diabetes model with insulin resistance. Methods Six-week-old rats were fed with high-fat and high-sugar 4 w, before with no food and water for 18 h, then intraperitoneally injected with 1% STZ solution (35mg . kg-1 ), without food and water for another 1 h;intraperitoneal injection with the same dose of citric acid-sodium citrate buffer was conducted in control group. Results Fourteen days after STZ injection, fasting blood glucose(FBG) levels kept stable, oral glucose tolerance test(OGTT) met the standards of diabetes, and fasting insulin levels were close to those in the control group. Mold rate reached 93% ,and death rate was zero. Conclusion Type 2 diabetes model was prepared to achieve the purpose of the experimental design.
