白英乙醇提取物诱导人胃癌SGC-7901细胞凋亡的实验研究

摘    要:目的:研究白英乙醇提取物对人胃癌SGC-7901细胞凋亡及凋亡相关基因Fas和caspase-3表达的影响。方法:体外培养人胃癌SGC-7901细胞;实验分正常对照、顺铂及白英乙醇提取物高、中、低剂量(10、5、2.5mg·mL-1)组。采用MTT法检测细胞增殖抑制率,荧光显微镜观察SGC-7901细胞凋亡及形态变化,半定量RT-PCR法分析基因Fas和caspase-3 mRNA的表达情况。结果:与正常对照组比较,白英乙醇提取物高、中、低剂量组对SGC-7901细胞增殖抑制均有促进作用(P<0.01),且呈明显的时效和量效关系,细胞凋亡显著增多,表现为细胞核固缩、染色质凝集、边聚等;基因Fas和caspase-3 mRNA表达显著升高(P<0.01或P<0.05),并随白英乙醇提取物浓度增加而加强。结论:白英乙醇提取物能剂量依赖性地抑制SGC-7901细胞增殖和诱导细胞凋亡,其作用可能与调控Fas和caspase-3 mRNA表达相关。

Abstract:OBJECTIVE: To explore the effect of ethanol extracts of Solanum lyratum on apoptosis of SGC-7901 cells and the mRNA expression of apoptosis associated genes Fas and caspase-3.METHODS: SGC-7901 Cells were cultured in vitro and randomly divided into the contral group, ethanol extracts of S. lyratum treatment groups(2.5 mg-mL-~, 5 mg. mL~, 10 mg-mL-1) and cisplatin group. The proliferation inhibitory rate was evaluated by MTT assay. Morphological changes and apoptosis of SGC- 7901 cells were observed under fluorescence microscope. The mRNA expression of Fas gene and caspase-3 were detected by semi-quantitive RT-PCR method. RESULTS: As compared with control group, the cell proliferation inhibitory rate and apoptosis rate of SGC-7901 cells were increased obviously in S. lyratum treatment groups (P〈0.01).Obvious morphological change of the SGC-7901 cells represented as the nuclear shrinkage, chromatin condensation and margination; the mRNA expression of Fas gene and caspase-3 were obviously increased as long as concentration of ethanol extract of S. lyratum increased(P〈0.01 ~P〈0.05). CONCLUSION: Ethanol extract of S. lyratum can dose-dependent induced apoptosis and inhibit proliferation, which are associated with regulating the mRNA expression of Fas gene and caspase-3.
