脊髓IL-33在关节炎大鼠痛觉过敏中的表达*

摘    要:目的 研究佐剂诱导关节炎大鼠脊髓组织中白细胞介素-33(IL-33)的变化,探讨IL-33在关节炎大鼠痛觉过敏中可能的作用。方法 50只成年雄性SD大鼠,体重200~250?g,随机分为对照组(Sham组)与佐剂性关节炎组(CFA组),每组25只。CFA组采用足底注射完全氟式佐剂(CFA)复制模型,术后第1、3、7、14及21天分别处死5只大鼠,观察各组大鼠行为学变化,检测机械痛阈和热痛阈,并采用Western blotting检测脊髓IL-33的表达。结果 CFA组大鼠自术后第3天起,各时间点的足趾和关节体积增大,与Sham组比较,差异有统计学意义(P?<0.05),CFA组术后6?h即出现机械痛阈和热痛阈下降,并持续至术后21天,与Sham组比较,差异有统计学意义(P?<0.05)。CFA组第3天开始出现脊髓IL-33的蛋白表达水平升高,并持续升高至术后21天,与Sham组比较,差异有统计学意义(P?<0.05)。结论 CFA组大鼠出现机械痛敏和热痛敏,脊髓IL-33表达增加,IL-33可能在类风湿关节炎的痛觉过敏中起重要作用。

Abstract:Objective To investigate the changes of interleukin-33 (IL-33) in spinal cord of rats with adjuvant arthritis and the possible role of IL-33 in hyperalgesia of arthritis. Method Fifty male SD rats, weighing 260 to 300 g, were randomly assigned to sham group and arthritis group, 25 in each group. Adjuvant arthritis group were induced by complete Freund''s adjuvant (CFA) injection. The behavioral changes of each group were observed. The mechanical pain threshold and thermal pain threshold were determined, and the expression of IL-33 in spinal cord was determined by western blotting at 1 d, 3d, 7 d, 14 d and 21 d after surgery.
