食管鳞状细胞癌的PTTG1表达及与上皮间质转化的关系及临床意义

摘    要:目的 分析食管鳞状细胞癌(ESCC)的垂体瘤转化基因1(PTTG1)表达及与上皮间质转化(EMT)的关系及临床意义。方法 选取2015年1月—2017年12月山东枣庄矿业集团中心医院接受手术治疗的73例经病理确诊的ESCC患者的手术标本,取癌旁组织作为对照。采用免疫组织化学法检测组织中E-钙黏附素、波形蛋白、N-钙黏附素及PTTG1表达,采用RNA干扰技术下调ESCC Eca109细胞系中PTTG1表达,采用Western blotting检测下调ESCC细胞系中E-钙黏附素、波形蛋白、N-钙黏附素及PTTG1表达,采用RT-PCR 检测下调ESCC Eca109细胞系中E-钙黏附素、波形蛋白、N-钙黏附素及PTTG1信使RNA(mRNA)表达。结果 与癌旁组织比较,癌组织的波形蛋白、N-钙黏附素及PTTG1阳性表达高(P?<0.05),E-钙黏附素阳性表达低(P?<0.05)。E-钙黏附素、波形蛋白、N-钙黏附素及PTTG1表达与ESCC患者的浸润深度、淋巴结转移、肝转移及分期相关(P?<0.05)。PTTG1表达还与ESCC患者的分化程度相关(P?<0.05)。PTTG1表达下调ESCC Eca109细胞系中E-钙黏附素蛋白及E-钙黏附素mRNA增高(P?<0.05),波形蛋白、N-钙黏附素、PTTG1蛋白及波形蛋白、N-钙黏附素、PTTG1 mRNA均降低(P?<0.05)。结论 ESCC肿瘤组织中存在PTTG1高表达,其表达与肿瘤的EMT相关。

Abstract:Objective To analyze expression of pituitary tumor-transforming gene 1 (PTTG1) in sophageal squamous cell carcinoma (ESCC) and the clinical significance in epithelial-mesenchymal transition (EMT). Methods A total of 73 ESCC patients who underwent surgery in our hospital from January 2015 to December 2017 were enrolled. Cancerous tissue as well as para-cancerous tissue were collected. Immunohistochemistry was used to detect the expression of E-cadherin, Vimentin, N-cadherin and PTTG1 in ESCC tissues.
