前列腺癌中HMBOX1的表达水平及其对细胞上皮间质化的影响*

摘    要:目的 探究HMBOX1在前列腺癌中的表达水平及其对前列腺癌上皮间质化的影响。方法 选取2015年10月—2017年10月在浙江省立同德医院确诊并行根治性前列腺切除术的68例前列腺癌患者作为研究对象(PC组),选取同期收治的70例良性前列腺增生患者作为对照(BPH组),采用免疫组织化学法检测HMBOX1在两组的表达;构建HMBOX1过表达(实验组)和对照组稳定细胞系,采用Western blotting检测HMBOX1、E-钙黏蛋白(E-cadherin)、波形蛋白(Vimentin)的表达;采用划痕实验检测HMBOX1过表达对细胞迁移的影响;采用Transwell检测HMBOX1过表达对细胞迁移能力的影响。结果 PC组HMBOX1蛋白的阳性表达率(19.12%)比BPH组(55.71%)下降(P?<0.05);实验组细胞中E-cadherin的表达水平高于对照组细胞(P?<0.05),Vimentin的表达水平较对照组细胞下降(P?<0.05);与对照组细胞比较,实验组细胞24?h后的划痕创口愈合面积较小,迁移速度降低(P?<0.05)。与对照组侵袭细胞数(421.88±45.70)个比较,实验组侵袭细胞数(172.06±19.25)降低(P?<0.05)。结论 HMBOX1在前列腺癌组织中表达降低,过表达HMBOX1可抑制前列腺癌的上皮间质化及肿瘤侵袭迁移能力。

Abstract:Objective To investigate the expression of HMBOX1 (Homeobox containing 1) in prostate cancer and its effect on epithelial-mesenchymal transition. Methods Totally 68 patients with prostate cancer who underwent concurrent radical prostatectomy in our hospital from October 2015 to October 2017 were selected as subjects (PC group), and 70 patients with benign prostatic hyperplasia admitted to the same period were selected as the control (BPH group); immunohistochemistry was used to detect the expression of HMBOX1 in PC and BPH groups; the HMBOX1 overexpression (experimental group) and control stable cell lines were constructed; western blot was used to detect the expression of HMBOX1, E-cadherin and Vimentin, and the wound-healing assay was used to detect the effect of HMBOX1 overexpression on cell migration; the effect of HMBOX1 overexpression on cell migration was analyzed by Transwell assay.
