大蒜油单独及与顺铂联合对人宫颈癌Hela细胞生长影响的研究

目的 :探讨大蒜油对人宫颈癌Hela细胞体外生长的抑制作用 ,以及大蒜油增加Hela细胞对顺铂的敏感性 ,以期对药物治疗宫颈癌以及其他肿瘤寻找更加完善的途径。方法 :体外培养人宫颈癌Hela细胞 ,分别用大蒜油、顺铂、大蒜油加顺铂处理 4 8小时 ,采用四甲基偶氮唑蓝 (MTT)比色法检测大蒜油和顺铂及两者联合应用对Hela细胞的生长抑制作用。结果 :大蒜油在浓度为 12 5mg/L、2 5mg/L、5 0mg/L、10 0mg/L时对Hela细胞的抑制率分别为 2 3 82 %、37 2 1%、5 2 2 5 %、6 7 6 9% ,与对照组 (加入等量培养液 )比较 ,差异有非常显著性 (P <0 0 1)。顺铂在浓度为 0 5mg/L、1 0mg/L、2 0mg/L时对Hela细胞的抑制率为 38 13%、4 6 16 %、6 0 38%。两者合用可大大提高对Hela细胞的抑制作用 ,抑制率分别为 4 2 5 0 %、5 5 38%、6 6 98%、78 31% ,与单纯应用大蒜油或顺铂比较差异有非常显著性 (P <0 0 1)。结论 :大蒜油能抑制Hela细胞的生长 ,同时能够增加Hela细胞对顺铂的敏感性。

Inhibition of Garlic Oil only or Combined with Cisplatin on Growth of Hela Cells

Objective: To investigate the inhibition of garlic oil to cervical cancer Hela cells in vitro and its sensitive effect on Hela cells to cisplatin, and to search for a perfect way in the treatment of cervical carcinoma with medicine. Methods: Hela cells cultured in vitro were treated with garlic oil only, Cisplatin only, garlic oil combined Cisplatin as long as 48 hours, and then were detected the inhibition of cells by MTT. Results: When the concentrations of garlic oil were 12.5 mg/L, 25mg/L, 50 mg/L, and 100 mg/L, the inhibition rate of the growth of Hela cells was 23.82%, 37.21%, 52.25%, and 67.69%, respectively, which there was significantly difference when compared with those of controls( P <0.01). On the concentration of 0.5mg/L, 1.0mg/L, and 2.0mg/L Cisplatin, the inhibition rate were 38.13%, 46.16%, and 60.38%, respectively. When combined with Cisplatin, the inhibition rate of garlic oil increased to 42.50%, 55.38%, 66.98%, and 78.31%, separatively, much higher than only garlic oil or Cisplatin used( P <0.01).Conclusions: Garlic oil can inhibits the growth of Hela cells as well as increase the sensitivity of Hela cells to Cisplatin.
