摘    要:观察了烟草特异亚硝胺NNK诱发HPV-18永生化人类支气管上皮细胞(BEP2D)恶性转化过程中细胞生物学特征的变化。500mg/LNNK处理2BEP2D细胞一天后,细胞在体外可连续传代,第5代细胞显示抗血清生长;第15代细胞在半固体琼脂中的集落形成率(0.038%)为对照细胞(0.0033%)的11倍;第25代细胞在裸鼠体内瘤,组织学证实为高分化鳞状细胞瘤,以上结果表明NNK致BEP2D细胞恶性转

Abstract:The malignant transformation of HPV-18 immortalized human bronchial epithelial cell(BEP2D) induced by tobacco-specific nitrosamine NNK and its biological characteristics were investigated. BEP2D cells treated with NNK at the concentration of 500 micrograms/ml for 24 hours could be subcultured continuously in vitro. The 15th passage cells in soft agar (0.038%) increased 11 fold compared with that of control cells (0.0033%). The 25th passage cells could grow into tumor in nude mice. The tumor was a squamous cell carcinoma in morphology confirmed by histopathological examination. The results indicated that the malignant transformation induced by NNK was a sequential process. This culture system provides a potential tool for the study of cell and molecular mechanism in the multistage carcinogenesis of human bronchial epithelial cells.
