1型糖尿病、2型糖尿病及2型糖尿病伴动脉硬化患者外周血白细胞端粒长度的研究

摘    要:目的:检测1型糖尿病(T1DM)、2型糖尿病(T2DM)、2型糖尿病伴动脉硬化(DAS)患者外周血白细胞端粒长度,分析糖尿病患者端粒长度变化的因素。方法:选择T1DM患者30例、T2DM患者60例、DAS患者40例和健康对照(NC组)40例,分别提取外周血白细胞,然后提取基因组DNA进行real-time PCR检测端粒长度。利用多元线性回归分析影响端粒长度变化的因素。结果:T1DM组、T2DM组和DAS组的端粒长度均小于NC组(P0.05);T1DM组端粒长度短于T2DM组和DAS组;DAS组较T2DM组更短。多元线性回归分析显示,T1DM组中,年龄与端粒长度呈负相关(P0.05);T2DM组中,年龄、体重指数(BMI)与端粒长度呈负相关(P0.05);DAS组中,患病时间、BMI与端粒长度呈负相关(P0.05)。结论:糖尿病患者的外周血白细胞端粒长度明显短于正常人,并且T1DM患者端粒长度短于T2DM;在2型糖尿病的对比中,DAS患者的端粒长度明显短于T2DM。患者的年龄、患病时间、BMI与端粒长度的缩短有密切的关系。

Abstract:AIM: To test the telomere length of peripheral blood leukocytes in the patients with type 1 diabetes mellitus (T1DM), type 2 diabetes mellitus (T2DM) and T2DM complicated with arterial sclerosis (DAS), and to explore the related mechanism. METHODS: Thirty patients with T1DM, 60 patients with T2DM, 40 patients with DAS and 40 normal controls (NC group) were enrolled in this trial and the peripheral blood leukocytes were isolated to extract DNA for determining telomere length by real-time PCR. Multiple linear regression analysis was performed to analyze the factors changing telomere length. RESULTS: The telomere lengths in T1DM group, T2DM group and DAS group were less than that in NC group (P < 0.05), that in T1DM group was shorter than those in T2DM group and DAS group, and that in DAS group was shorter than that in T2DM group. The results of multivariate linear regression analysis showed that age was negatively correlated with telomere length (P < 0.05) in T1DM group, age and body mass index (BMI) were negatively correlated with telomere length (P < 0.05) in T2DM group, and duration of illness and BMI were negatively correlated with telomere length (P < 0.05) in DAS group. CONCLUSION: The telomere length of peripheral blood leukocytes in diabetic patients is shorter than that in normal people, and the telomere length of T1DM patients is shorter than those of T2DM and DAS patients. The telomere length of DAS patients is shorter than that of T2DM patients. The telomere length shortening in diabetic patients is mainly associated with age, duration of illness, and BMI.
