激活骨髓和淋巴因子激活的杀伤细胞生物活性的实验研究

摘    要:目的 :证实激活骨髓 (Activated bone marrow,ABM)具有与淋巴因子激活的杀伤 (LAK)细胞不同的生物学效应。方法 :应用重组白细胞介素 - 2 (r IL - 2 )、抗 CD3 单抗体外激活骨髓和外周血单个核细胞 ,以 MTT比色分析法测定 ABM、L AK细胞的杀伤活性 ,采用甲基纤维素半固体培养法进行粒 -巨噬细胞系祖细胞 (CFU- GM)、红系祖细胞 (BFU- E)培养。结果 :r IL- 2 +抗 CD3 单抗激活的 ABM组杀伤活性最强 (6 5 .8± 9.2 ) % ,r IL - 2激活的 ABM组次之为 (5 6 .3± 7.9) % ,活性强于相同条件下的 L AK组 ,两组差异具有显著性 (P<0 .0 1)。增殖倍数与相同条件下的 LAK细胞相比差异亦具有显著性。体外激活 3~ 5 d的 ABM与同期培养的未激活的骨髓细胞相比 BFU- E、CFU- GM形成率差异无显著性 (P>0 .0 5 ) ,LAK细胞则不具有造血祖细胞活性。结论 :ABM的杀伤活性和增殖效应明显强于 L AK细胞 ,且能保持造血祖细胞活性 ,抗 CD3 单抗能够促进 r IL - 2诱导的 ABM抗肿瘤活性和增殖效应 ,不损伤造血祖细胞活性 ,是优于 LAK细胞的过继免疫疗法。

Abstract:Objective:To prove that activated bone marrow (ABM) has different biological effect different from LAK cell. Methods: Anti CD 3 monoclonal antibody and rIL 2 were used to activate mononuclear cells obtained from bone marrow and blood and to test the killing activety of ABM and LAK cell;CFU GM and BFU E were cultivated with methylcellulose semi solid culture method. Results:rIL 2+anti CD 3 monoclonal antibody had the largest activity;the percentage of killing activity of ABM was (65 8±9 2),and it was (56 3±7 9) by rIL 2 . ABM had large killing activity than LAK cell;there were obvious differences( P <0.01) . Compared to IL 2,the cell proliferation after 7 days culturing had obviously different. There was no obvious difference in CFU GM,BFU E formation rate between ABM that was activated by marrow cell for 3~5 days amd the bone marrow cell that was not activated but cultured for same time. Conclusion:Compared to IL 2,ABM had larger killing activity and proliferate effect,and can keeping activity of progenitor cell. Anti CD 3 monoclonal antibody can enhance the killing effect and the proliferation of ABM induced by rIL 2, producing no injury of activity of progenitor cell.It is better immunotherapy than LAK cell.
