大鼠心肺复苏后脑组织基质金属蛋白酶及其组织抑制剂表达的研究

摘    要:目的探讨心肺复苏早期脑组织基质金属蛋白酶-2(MMP-2)、MMP-9及基质金属蛋白酶组织抑制剂-1(TIMP-1)的mRNA表达变化。方法用窒息法建立大鼠心肺复苏模型。80只SD大鼠随机分为假手术对照组和复苏组,然后依据时间分为假手术或复苏自主循环恢复(ROSC)后即刻及0.5、3、6和9h组。检测各组大鼠脑组织MMP-2、MMP-9及TIMP-1的mRNA表达情况。结果心肺复苏后3h大鼠脑组织MMP-9及TIMP-1的mRNA表达水平均开始上升,6h时显著增高,MMP-9/TIMP-1比值也相应增大。MMP-2 mRNA水平在心肺复苏后9h内未见明显升高。结论心肺复苏后早期就出现MMP-9、TIMP-1的mRNA表达增加及比例失衡,而MMP-2 mRNA水平在早期无明显变化。

Abstract:Objective To study the change in mRNA expression of matrix metalloproteinase (MMPs) and tissue inhibitor1 of matrix metalloproteinase (TIMP1) in brain after cardiopulmonary resuscitation (CPR) for asphyxial cardiac arrest in rat. Methods The animal model of cardiac arrest was reproduced by clamping endotracheal tube at the expiration. Eighty male SD rats were randomly divided into two groups: control group and resuscitation group, and they were again divided into 0, 0.5, 3, 6, 9 hours subgroups ( n =8) . Evans blue content and mRNA expressions of MMPs and TIMP1 in the brain after CPR were determined respectively. Results The mRNA expression of MMP9 and TIMP1 was upregulated 3 hours after restoration of spontaneous circulation (ROSC). At 6 hours after ROSC, they were markedly high , and the ratio of MMP9/ TIMP1 was higher too. The mRNA expression of MMP2 showed no significant change at 9 hours after ROSC. Conclusion The mRNA expression of MMP9 and TIMP1 and MMP9/TIMP1 are increased at early stage after CPR, but the mRNA expression of MMP2 shows no significant change.
