肾移植患者术后外周血CD4+CD25+调节性T细胞的检测及其与瘦素水平的相关性

目的:了解肾移植患者术后外周血中CD4 CD25 调节性T细胞的比例和瘦素水平的变化及其临床意义,探讨CD4 CD25 调节性T细胞与瘦素水平的相关性。方法:应用放射免疫方法检肾移植患者及健康对照者外周血中瘦素的水平,流式细胞术检测外周血中CD4 CD25 调节性T细胞占CD4 T细胞的比例。结果:移植近期组(1年)与对照组患者血浆中瘦素浓度比较无差异,但移植远期组的瘦素浓度(2.5年)高于对照组和移植近期组。且外周血中CD4 CD25 调节性T细胞的比例与瘦素呈显著负相关(r=-0.83,P<0.01)。结论:肾移植术后的远期高瘦素血症可能对CD4 CD25 调节性T细胞具有负性调节作用,也许不利于移植耐受的维持。

Observation of peripheral blood CD4+CD25+ regulatory T cells and analysis of correlation between its proportion and the level of Leptin in posttransplant recipiens

Objective:To evaluate the clinical significance of leptin levels and the proportion of CD4 CD25 regulatory T cells in patient with renal transplantation, to investigate the correlation between its proportion and the level of leptin.Methods:Radio immunoassay was used to evaluate plasma leptin level and Flow cytometry was used to detect the proportion f CD4 CD25 regulatory T cells in patient with renal transplantation.Results:The level of Leptin measured in kidney transplants patients indicated that there was no significant difference in Leptin level between the control and short term (1 year) posttransplant recipients. But the level of Leptin was higher on long term (2.5 years) posttransplant recipients. The CD4 CD25 T cells population in posttransplant recipiens was negatively correlated with leptin level(r=-0.83 , P<0.01).Conclusions:The high level of leptin in posttransplant recipiens is correlated with the down-regulation of CD4 CD25 regulatory T cells, and may not profitable for the maintenance of transplantation tolerance.
