IL-10基因-627C/A多态性与儿童哮喘病之间的相关性研究

目的 研究白介素10(IL-10)基因-627C/A多态性与哮喘病之间的相关性.方法 对110例哮喘患儿和63位正常志愿者进行血液IL-10浓度的检测、IL-10基因-627C/A多态性分析.结果 IL-10实验的准确度95.64%,精密度95.46%,均达到了试剂盒的要求.哮喘组血清IL-10质量浓度(99.78±34.01)pg·ml-1,对照组血清IL-10质量浓度(82.38±48.91)pg·ml-1,差异具有统计学意义(P＜0.05).哮喘组中基因型分布为:AA型20.9%,AG+AA型79.1%;对照组中分布为:GG型63.5%,AG+AA型36.5%.哮喘组中等位基因频率A和G分别为56.3%和43.7%;对照组中分别为66.9%和33.1%.基因型频率和等位基因频率在两组间分布差异有统计学意义(P<0.05).结论 哮喘患儿和正常者IL-10的分泌量不同.IL-10基因-627位点单核苷酸多态性(SNP)可能与哮喘的发生相关;627 GG可能增加哮喘发生的风险.

Study of the correlation between IL-10 gene-627C/A polymorphism and childhood asthma

Objective: To Study the correlation between IL-10 gene-627C/A polymorphism and childhood asthma.Methods: 110 asthmatic children,and 63 normal volunteers,blood levels of IL-10 detection,IL-10 gene-627C/A polymorphism were tested and analyzed.Results: IL-10 experimental accuracy was 95.64%,95.46%,both reached the kit requirement.The asthma group had serum IL-10 levels(99.78±34.01) pg·ml-1 while the control group had serum IL-10 level(82.38±48.91) pg·ml-1,with a statistically significant difference(P<0.05).Genotype distribution for the asthmatic group was that AA-type accounted 20.9%,AG+AA-type 79.1%;while in the control group,GG-type 63.5%,AG+AA-type 36.5%.Asthma group A and G allele frequencies were 56.3% and 43.7%;the control group were 66.9% and 33.1%.Statistical analysis showed that genotype and allele frequency distribution differences between the two groups were statistically significant(P<0.05).Conclusion: The secretion of IL-10 between asthma and normal children is different.IL-10 gene-627 single nucleotide polymorphism(SNP) may be associated with the occurrence of asthma.627 GG may increase the risk of asthma.
