人源肝细胞系CL-1、HepFMMU、HepG2、C3A生物学特性与功能的比较

摘    要:目的:比较体外培养的四种人源肝细胞系CL-1、HepFMMU、HepG2、C3A生物学与功能特性的异同,初步评价其应用于生物人工肝的可行性,为其在组织工程学研究和临床医学中的应用奠定基础。方法:4种肝细胞在接种密度相同的条件下培养7d,观察4种细胞形态特征和功能状态,检测培养液中白蛋白、尿素及安定代谢量。结果:4种肝细胞系均具有较快的生长速度,功能学比较C3A具有良好的白蛋白合成和安定代谢功能;CL-1具有良好的尿素合成功能。结论:4种肝细胞具有良好的增殖能力,C3A具有良好的白蛋白合成及安定代谢能力,但其尿素合成能力较差,需进一步改进;CL-1具有较好的尿素合成能力,但其安定代谢和白蛋白合成功能较差;HepFMMU功能较差,不适宜用于生物人工肝。

Abstract:Objective To evaluate the potential feasibility of using of human hepatocyte lines in bioartificial liver and tissue engineering by comparing their biological features and functions.Methods Four hepatocyte lines were cultured for seven days under the condition of the same density of the cells at the beginning,the characteristics of cell morphological were observed,the synthesis level of albumin,biotransformation of urea and the diazepam metabolism level were detected.Results Four human hepatocyte lines have same speed of growth,C3A has the best capacity of albumin synthesis and the diazepam metabolism;the urea concentration of CL-1 was the best among four hepatocytes.Conclusion Four hepatocyte lines have better capacity of proliferation;C3A has the best capacity of albumin synthesis and the diazepam metabolism,however it’s biotransformation of urea was poor and need further improvement;CL-1 has the best urea biotransformation ability,but it’s capacity of albumin synthesis and the diazepam metabolism was not good;the liver-specific function of HepFMMU was poor and not appropriate for bioartificial liver.
