聚乙烯醇水凝胶对成骨细胞毒性的初步研究

目的：研究聚乙烯醇水凝胶与成骨细胞的生物相容性，为聚乙烯醇（ Polyvinyl Alcohol，PVA）水凝胶应用于口腔软组织增量提供实验依据。方法：按照选定实验条件制备出PVA水凝胶块，将分切成合适大小的PVA水凝胶直接与成骨细胞（ OB）联合培养，观察细胞的生长增殖情况，用MTT法检测PVA水凝胶对成骨细胞的毒性。结果：与PVA水凝胶直接接触的成骨细胞第1、2 d的相对增殖率接近99％，毒性分级为1级；第5 d的相对增殖率在99％以上，毒性分级为0级；第8、10 d的相对增殖率在90％以上，毒性分级为1级。 PVA水凝胶组与阴性对照组间吸光度值无统计学差异（P＞0．05），而PVA水凝胶组与空白对照组及阳性对照组的吸光度值比较，均有显著差异（P＜0．05）。结论：PVA水凝胶凝胶具有良好的生物相容性，可用于口腔软组织增量。

A preliminary study the toxicity of polyvinyl alcohol hydrogel on osteoblasts

Objective:To study the biocompatibility of polyvinyl alcohol( PVA) hydrogel with osteoblasts and provide experimental evidence for PVA hydrogel that applied to the oral soft tissue increment.Methods:According to the experimen-tal conditions several pieces of PVA hydrogel was prepared which was cutted into the appropriate size for cultured with osteo-blasts( OB) directly.The cell proliferation and the toxicity of PVA hydrogel to osteoblasts by MTT assay was observed.Re-sults:Directly contact with the PVA hydrogel,the relative growth rate of osteoblasts was closed to 99%in 1,2 d with toxicity rating level 1,more than 99%in 5 d with toxicity rating level 0 and in 8,10 d with toxicity rating level 1.There was no statis-tically significant difference in the absorbance values between PVA hydrogel group and negative control group(P>0.05). There were statistically significant differences in the absorbance values between PVA hydrogel group and control group or pos-itive control group,respectively(P<0.05).Conclusion:PVA hydrogel with satisfying biocompatibility can be used for oral soft tissue increment.
