针刺对舌癌变模型大鼠血清CD4+和CD8+的影响研究

目的:探讨针刺对口腔黏膜癌变模型大鼠细胞免疫功能的影响。方法:利用0.002%的4-硝基喹啉-1-氧化物(4NQO)饮水法建立大鼠舌癌变模型,于建模13～18周给予针刺治疗,以阳性药物维甲酸作对照,采用HE染色观察大鼠舌黏膜组织形态的变化,运用酶联免疫法检测血清CD4+、CD8+的含量。结果:组织病理学显示,模型组非典型增生和癌变程度最重、药物组次之、针刺组最轻。各组血清CD4+含量与空白组比较均有升高,且针刺组升高明显(P<0.05);针刺组CD8+含量和CD4+/CD8+比值分别均较模型组和药物组有明显升高(P<0.05)和降低,且与空白组最接近。结论:针刺在口腔粘膜癌变的发展进程中显示一定的阻抑作用,可能与其调节大鼠细胞免疫功能有一定关联。

Effects on blood serum CD4+ and CD8+ intervened by acupuncture on tongue cancer rats model

Objective： To explore the effects of immune function of tongue cancer rats model intervened by acupuncture. Method： Using 0.002 % 4-Nitroquinoline 1-oxide （4NQO） in drinking water to establish the rats model for oral mu- cosa carcinogenesis, acupuncture was implemented in 13-18 weeks, positive controlled by retinoic acid,to observe the alter- ation of oral mucosa morphology by HE stain and the quantity of blood serum CD4＋ and CD8＋ by Enzyme linked immunosor- bent assays （ELISA）. Result： Histopathology makes it clear that atypical hyperplasia and Cancerous degree of model group is the most serious one, medicine group takes second place, and the destructiveness of acupuncture group is the lowest. Con- trast to black control group, Serum CD4＋ in other groups increase, in which the acupuncture group is remarkable,making a significant difference （P 〈0.05）： both serum CD8＋and CD4＋ / CD8＋ ratio of acupuncture group increases （P 〈0.05）, and decres respectively more than model group and medicine group, closing to black group. Conclusion： To a certain extent, it is obvi- ously illustrated （P 〈0.05） and decrease respectively that there is inhibition effect of acupuncture in the occurrence and de- velopment of oral mucosa carcinogenesis, relevance to rats immune function regulation probably.
