人血清IgG类抗LDH-C4抗体的ELISA检测及其意义

目的建立检测人血清IgG类抗精子特异性乳酸脱氢酶(LDH-C4)抗体的ELISA法,探讨人血清抗LDH-C4抗体与免疫性不育的关系。方法以重组人LDH-C4(rhLDH-C4)作为包被抗原,建立人血清IgG类抗LDH-C4抗体检测的间接ELISA方法。应用该方法检测74例健康人群(正常生育组)和177例不育人群(不育组)血清IgG类抗LDH-C4抗体。结果初步建立了检测人血清IgG类抗LDH-C4抗体的间接ELISA方法。不育组血清IgG类抗LDH-C4抗体阳性率(30.51%)显著高于正常生育组(9.46%)(P0.01),女性不育组阳性率(31.46%)与男性不育组(29.55%)差异不显著(P0.05)。结论人血清中存在着IgG类抗LDH-C4抗体,此类抗体可能与免疫性不育有密切关系。

ELISA method for detection of IgG-type anti-sperm specific lactate dehydrogenase antibody in human sera and its clinical significance

Objective To establish an ELISA method for the detection of IgG-type anti-sperm specific lactatedehydrogenase antibody in human sera and investigate the relationship between anti-sperm specific lactate dehydrogenase antibody and immune infertility.Methods An indirect ELISA was established using recombinant human sperm-specific dehydrogenase（rhLDH-C4） antigen for plate coating and anti-LDH-C4 antibody（IgG type） in the sera from 74 fertile individuals（fertile group） and 177 unexplained infertile patients（infertile group） was detected.Results An indirect ELISA method for detecting IgG type antiLDH-C4 antibody in human sera was established.Of 177 infertile patients（in infertile group）,54 were found positive for serum IgG type anti-LDH-C4 antibody,resulting in a positive ratio of 30.51%（54/177）,which was significantly higher than that in fertile group（7/74 or 9.46%,P0.01）.Within the infertile group,no significant difference of positive ratios were observed between female and male members（31.46% vs 29.55%,P0.05）.Conclusion There was IgG-type anti-LDH-C4 antibody in human sera,and this antibody might be closely related to infertility.
