中药抗炎作用与对大鼠腹腔巨噬细胞产生白三烯B_4及5-HETE的影响

摘    要:本文建立了提取与测定大鼠腹腔巨噬细胞(PMφ)生物合成白三烯B_4(LTB_4)及5-羟基二十碳四烯酸(5-HETE)的高效液相色谱法,并研究了4个单味中药抗炎作用与对PMφ产生LTB_4及5-HETE的影响。结果表明大黄、厚朴、白头翁及败酱草提取物对大鼠PMφ合成以上二种介质均有明显抑制作用。对炎性介质LTB_4产生的抑制率(%)分别为:大黄素(终浓度取5.0×10~(-6)mol/L)为88.0%;厚朴为99.8%;白头翁为94.9%及败酱草为57.9%(以上终浓度均取5.9×10~(-2)g/L)。

Abstract:LTB4 and 5-HETE were determined by High Performance Liquid Chromatography,The anti-inflammatory action of four extractions from Chinese herbs including Emodin,Magnolia Officinalis,Pulsatilla Chinensis and patrinia vilosa,was investigated for its effect on the synthesis of LTB4 and 5-HETE released form peritoneal macrophages stimulated by calcium ionophore A23187,The results showed that the four herb extractions significantly inhibited the biosynthesis of both LTB4 and 5-HETE,the inhibite rate was from 60%to 98%as same as that of flufenamic acid.We suggested that the mechanism of antiinflammatory action of these agents may be related to the suppresion of lipoxygenase.
