富马酸二甲酯的致畸性研究

摘    要:将妊娠大鼠95只,分为六组,经口灌入DMF,剂量分别为0.0、89.0、178.0、356.0和508.0mg/kg体重,另设一个阳性对照(敌枯双)组。自妊娠第6~15天染毒,每天一次。结果表明DMF具有胚胎毒性,当剂量达598.0mg/kg体重时,胎鼠全部死亡,胚胎大部分吸收。剂量达356.0mg/kg时,死胎率显著增加。剂量为178.0mg/kg时,引起胚胎吸收率显著增加。DMF未引起任何胎鼠外观和内脏畸形,亦未见骨骼畸形,但剂量达356.0mg/kg时,引起3只胎鼠的头骨和尾骨骨化迟缓。阳性对照敌枯双引起胎鼠典型的外观、内脏和骨骼畸形。

Abstract:95 pregnant rats were divided into 6 groups. Dose of dimethyl fumarate(DMF) for each group was 0.0, 89.0, 178.0, 366.0 and 508.0mg/kg-BW, respectively. In addition, the positive control group was given N,N'-methylenebis(2-amino-1,3,4-thiadiazole). The duration of dosing was from day 6 to day 15 of pregnancy. The result showed that DMF appeared embryotoxicity. 508.0 mg DMF/KgBW induced death of fetuses and embryo absorption. The absorption rate increased significantly in the group of 178.0mg DMF/KgBW. Death rate of fetus obviously increased in the group of 356.0mg DMF/KgBWN.o malformation in the organs and bone was observed in the test groups. Delay of ossification in the coccyx and skull bone was found in 3 rats of the group of 356.0mg DMF/KgBW.
