微量元素铁与肿瘤发生及发展的关系

目的 铁既是生命所必须的,但又与肿瘤的发生、发展密切相关,其机制并不明确.本文旨在总结微量元素铁与肿瘤发生、增殖、转移的关系及其机制.方法 应用PubMed及CNKI期刊全文数据库检索系统,以“铁,肿瘤”为关键词,检索1990-2016年的相关文献.纳入标准:(1)细胞实验、动物模型、人群研究中微量元素铁与肿瘤发生、发展的关系;(2)氧化应激损伤、炎症反应、免疫应答、信号通路异常在微量元素铁与肿瘤关系中的作用.根据纳入标准,符合分析的文献共70篇.结果 大量研究发现,铁过载与肿瘤的发生及发展密切相关,铁通过诱导氧化损伤、促进炎症因子的产生、降低免疫系统对肿瘤的监视等机制增加肿瘤的发病风险,铁还可以改变正常信号转导通路促进肿瘤增殖与转移.最近这些年许多参与铁代谢的蛋白被发现,靶向铁的治疗可能作为肿瘤治疗的新策略,但是其需要更多的临床资料来证实.结论 通过对铁元素与肿瘤发生及发展关系的研究,能够给肿瘤的预防及治疗提供新的思路与方法.

Relationship between iron and the tumorigenesis as well as tumor development

OBJECTIVE Iron is necessary for life,but also closely related to tumorigenesis and tumor development,its mechanism is not clear.This review was to summary the relationship between iron and the occurrence,proliferation,metastasis of cancer,and its mechanism.METHODS The papers published from 1990 to 2016 were retrieved with key words "iron,cancer" in PubMed and CNKI periodical full text database retrieval system.Inclusion criteria:(1) the relationship between iron and the occurrence,development of cancer in cell experiments,animal models,population studies.(2) the effect that oxidative stress,inflammation,immune response,abnormal signaling pathways played in iron and cancer.According to the inclusion criteria,70 literatures were collected.RESULTS Numerous studies found that iron overload is closely related to tumorigenesis.Iron overload promote tumorigenesis by inducing oxidative damage and inflammatory cytokine production,reducing the immune surveillance of cancer,iron can also alter the normal signal transduction pathways to promote tumor growth and metastasis.Many of the proteins involved in iron metabolism were found in recent years.The treatment targeting iron may be a new therapy of cancer,but needing much more clinical data to verify.CONCLUSION To study the relationship between iron and tumorigenesis,it is possible to provide new ideas and methods for cancer prevention and treatment.
