茶树(Camellia Sisensis O. Ktze)叶片培养的器官发生

摘    要:以叶片为外植体,采用MS基本培养基,附加1~5mg/L的6-苄基嘌呤(6-BA)、2mg/L吲哚乙酸(IAA)、3mg/L赤霉素(GA3),0.2~0.3%活性炭诱导愈伤组织。并用MS或ER为基本培养基,附加0.5~5mg/L6-苄基嘌呤(6-BA),0.5mg/L吲哚丁酸(IBA)诱导胚状体和器官,进一步形成小植株。采取MS加5~8mg/L细胞分裂素和0.5~1mg/L的细胞生长素,每天光照10小时便形成“丛生叶”,植株矮小;若上列激素的配比接近时,每天光照12~14h,即打破丛生叶形态,节间抽长形成正常的互生叶小植株。

Abstract:n this study, leaves of Camellia Sinensis O. Ktze. were cultured as explants, and thecallus was obtained in MS medium supplemented with 6 - BA (1 - 5mg/L) , IAA(2mg/L. GA3 (3mg,L) and activated carbon (0.2-0.3%). Embryogenesis and organogenesiswere induced in the MS or ER medium added with 6-BA (0.5-5.omg/L), IBA (0.5-5.0ml/L)and finally complete plantlets were formed. Caespitose leaves were formed in the MSmedium supplemented with cytokin (5-8mg/L) and auxin (0.5-l.0mg/L) under 10 hr dailyphotoperiod of l, 500 lux light intensity, and the plantlets were small and short. If theproportion of auxin to cytokin is close to l: l, the caespitose state was broken under12-14 hr daily photoperiod of 2, 000 lux light intensity, the internodeswere stretchedand the leaflets with alternate leaves were formed.
