补肾疏肝法调节去势大鼠血清E2水平及下丘脑5-HT含量的机理研究

目的:研究针刺对去势大鼠血清雌二醇及下丘脑5-羟色胺含量的影响,观察针刺前后血清雌二醇及下丘脑5-羟色胺含量的变化。方法:将30只实验大鼠按体质量随机分为4组,针刺组及雌激素组分别10只,模型组及对照组分别5只。对照组(A组),模型组(B),针刺组(C),雌激素组(D)。A组行假手术,其余3组大鼠行卵巢摘除术,术后1周后开始给药。A、B组分别给予生理盐水2mL/d灌胃。C组给予针刺穴位(合谷、足三里、太冲、内关、三阴交、关元、气海),隔日1次;D组给予雌二醇(0.20mg/kg)肌内注射,隔日1次。针刺及给药30次。在末次治疗24h后,经腹主动脉采血,颈椎脱臼法处死大鼠,冰盒上迅速分离下丘脑,采用化学发光法检测血清雌二醇的含量及固相夹心法酶联免疫吸附试验(ELISA)测定下丘脑5-HT含量。结果:模型组血清E2、下丘脑5-HT均降低,符合围绝经期抑郁症的实验室指标变化。针刺和雌激素均有升血清E2的作用,二者之间比较无统计学意义,但是针刺升血清E2作用弱于雌激素组;针刺与雌激素均影响下丘脑5-HT含量,但是针刺影响5-HT含量的作用较雌激素弱。5-羟色胺的含量与血清中雌二醇的存在正相关性。结论:采用补肾疏肝法针刺穴位,以滋阴补肾、疏肝解郁、调理冲任,调节去势大鼠内分泌功能,影响去势大鼠神经系统的功能状态。

Mechanism Study of Kidney-Tonifying-And-Liver-Soothing Therapy on Regulating Levels of Estradiol And 5-HT in Ovariectomized Rats

Objective:To study the effect of estradiol and 5-HT by acupuncture some acupoints of neutered big rats;and to research the changing of estradiol and 5-HT.Method:The 32 rats according to body weight were randomly divided into four groups:normal control group(A),Model group(B),acupuncture treatment group(C)and estrogen treatment group(D).In addition to A group,the other three groups were ovariectomized.Administration after a week,A and B groups were given normal saline gavage;C group was given acupuncture every other day;D group received estradiol intramuscuLar injection every other day.Acupuncture and administration were given 30 times.Detect serum estradiol levels by Chemiluminescent and test 5-HT content in Hypothalamic by ELISA.Result:The levels of serum E2 and hypothalamic 5-HT of model group decreased,accord with laboratory indices changes of perimenopausal depression.Acupuncture and estrogen have the role to raise serum E2.But the role of acupuncture is lower than estrogen.There is positive correlation between serum E2 and hypothalamic 5-HT.Conclusion:Acupuncture points can smooth the liver and reinforce rssence of the kidney,conditioning Chong and Ren,regulating peri-menopausal period of serum E2 levels in patients with depression,and regulate Hypothalamic-pituitary-ovarian axis.Reconstructing part of the estrogen negative feedback influenced neutered big rats of the nervous system functional state.
