缬沙坦对糖尿病大鼠肾脏肥大及P27表达的影响

摘    要:目的:探讨血管肾张素受体I拮抗剂(ATIRa)对糖尿病肾脏肥大及细胞周期蛋白P^27表达的影响。方法:大鼠腹腔注射链尿佐菌素(STZ)诱发糖尿病模型,每日灌胃给予ATIRa缬沙坦(8mg/kg),共2周。采用图像分析仪测量肾小球和肾小管直径并结合肾重/体质量比评价肾脏肥大状态,流式细胞仪定量检测肾皮质P^27表达。结果:糖尿病组较对照组肾小球、肾小管直径增大,肾重/体质量比增加、P^27蛋白表达增强;缬沙坦治疗组较糖尿病组肾脏肥大减轻、P^27表达减弱。结论:缬沙坦可能通过影响细胞周期蛋白P^27表达而抑制肾脏肥大,从而发挥肾脏保护作用。

Abstract:Objective To investigate the effects of valsartan on renal hypertrophy and the expression of p27 in the kidney of diabetic rats. Methods Diabetes was induced in 18 SD rats by intraperitoneal injection of STZ(65 mg/kg). Valsartan(40 mg/kg) was delivered daily by gavage from the next day of the induction to diabetes for 2 weeks. TD 2000 image pattern analysis system was used to detect diameters of each glomerular and tubular cross section; kidney/body weight ratio was also used as an index to evaluate renal hypertrophy. The expression of P27 was detected by flow cytometry. Results Diameters of glomerular and tubular cross section, kidney/body weight ratio and the expression of p27 increased in the diabetic kidneys(P < 0.05 ). Above alterations were ameliorated by valsartan treatment (P < 0. 05). Conclusion The protective effect of valsartan on diabetic kidney may be partly through inhibiting renal hypertrophy by regulating the expression of cell cycle related protein P27.
