款冬花紫菀配伍对绿原酸在大鼠体内药代动力学影响

摘    要:目的:研究款冬花、紫菀配伍对绿原酸在大鼠体内药代动力学参数的影响。方法:在筛选款冬花止咳有效部位、紫菀止咳增效部位的基础上,将SD大鼠随机分成4组,分别灌胃给予绿原酸单体、款冬花部位、紫菀部位、款冬花部位+紫菀部位(即部位配伍),采用HPLC法测定血浆中的绿原酸浓度,并用WinNonlin药动学软件根据非房室模型法计算药动学参数。结果:大鼠灌胃绿原酸单体、款冬花部位、紫菀部位、部位配伍后的主要药动参数:AUC0-t分别为245.09±14.05,349.27±62.15,105.11±18.57,496.77±77.75μg·mL^-1·min^-1;t1/2分别为323.70±51.00,464.53±126.10,408.27±31.30,203.02±7.12min;Cmax分别为1.05±0.13,1.52±0.55,0.63±0.04,4.49±0.59μg·mL^-1;Tmax分别为60.00±0.00,15.00±0.00,15.00±0.00,15.00±0.00min。结论:紫菀能促进大鼠对款冬花中绿原酸的吸收,加快绿原酸的消除,减少绿原酸在体内的蓄积,可以部分诠释款冬花、紫菀“相须配伍”增效的机理。

Abstract:Objective:To study the pharmacokinetic features of chlorogenic acid before and after the combination of the extracts of Farfarae Flos and Asteris Radix in rats. Methods : SD rats were randomized into 4 groups and were intragastric administered with chlorogenic acid (CA) , Farfarae Flos extract (FF), Asteris Radix extract (AR) and the combined extract of FF+AR, respectively. A HPLC method was established to determine chlorogenic acid in rat plasma. The main pharmacokinetic parameters were calculated with WinNonlin software by non-compartmental methods. Results: The main pharmacokinetic parameters of chlorogenic acid in 4 groups of CA, FF, AR and FF+AR were as follows in order: AUC (μg·mL^-1·min^-1) were 245.09± 14.05, 349.27±62. 15, 105.11±18.57, 496.77±77.75; t1/2 (min) were 323.70±51.00, 464. 53±126.10, 408.27±31.30, 203.02±7.12; Cmax (μg·mL^-1) were 1.05±0.13, 1.52±0.55, 0.63±0.04, 4.49±0.59; Tmax (min) were 60. 00±0.00, 15.00±0.00, 15.00±0.00, 15.00±0.00. Conclusion : The synergy of compatibility of Farfarae Flos and Asteris Radix could be explained by enhancing the absorption and accelerating the elimination of chlorogenic acid in rat plasma.
