高迁移率族蛋白B1 在类风湿关节炎中的表达水平研究

目的 探讨高迁移率族蛋白B1（HMGB1）在类风湿关节炎中的表达。方法 选取2017 年5 月-2018 年5 月在吉林大学第一医院行关节置换术或滑膜清理术的类风湿关节炎患者49 例作为研究组；另选取该院同期行手术治疗的骨关节炎患者30 例作为对照组。所有研究对象入院后第3 天清晨空腹采集外周静脉血，分离血清，采用酶联免疫吸附法测定HMGB1 含量；取保存的所有研究对象的病损关节滑膜组织石蜡病理切片标本，采用链霉菌抗生物素蛋白- 过氧化物酶连结（SP）法检测HMGB1 的表达。结果 研究组血清HMGB1 水平高于对照组（P <0.05）。研究组HMGB1 阳性表达率高于对照组（P <0.05）。研究组SP 法染色评分高于对照组（P <0.05）。结论 HMGB1 在类风湿关节炎中高表达，其可能在类风湿关节炎组织损伤发生、发展中发挥重要作用，可作为类风湿关节炎辅助诊断指标之一。

Expression of HMGB1 in rheumatoid arthritis

Objective To investigate the expression of high mobility group protein B1 (HMGB1) in rheumatoidarthritis. Methods Forty-nine patients with rheumatoid arthritis who underwent joint replacement or synovectomyin the First Hospital of Jilin University from May 2017 to May 2018 were selected as the subjects. In addition,30 patients with osteoarthritis who underwent surgery in the same hospital during the same period were selectedas the control group. The peripheral venous blood was collected in the morning of the 3rd day after admissionand the serum was separated. The serum content of HMGB1 was determined by enzyme-linked immunosorbentassay. And the expression of HMGB1 in the diseased synovial tissues was detected by immunohistochemical SPmethod. Results The serum HMGB1 level of the study group was higher than that of the control group (P < 0.05).The positive expression rate of HMGB1 in the study group was higher than that in the control group (P < 0.05).The score of immunohistochemical SP staining in the study group was higher than that in the control group(P < 0.05). Conclusions The high expression of HMGB1 in rheumatoid arthritis may play an important role in thepathogenesis of tissue damage in rheumatoid arthritis, and HMGB1 may be used as an auxiliary index in the diagnosisand treatment of rheumatoid arthritis.
