摘    要:目的:优选苦参素缓释微球制备工艺。方法:采用正交试验法,以微球载药量和包封率为指标,考察壳聚糖质量分数、苦参素与壳聚糖质量比、戊二醛用量和交联时间对制备工艺的影响。结果:最佳制备工艺为壳聚糖质量分数2%、苦参素与壳聚糖质量比1∶1、戊二醛用量1 mL和交联时间2 h,载药量和包封率分别为12.56%和41.98%。结论:正交试验法优选苦参素缓释微球的制备工艺简便、快速、准确。

Abstract:Objective To optimize the preparation method of oxymatrine sustained-release microspheres.Methods The orthogonal design was used to test the influence of four main factors in preparation,including amount of chitosan,mass ratio of oxymatrine and chitosan,amount of glutaraldehyde and emulsion cross-linking time.The drug loading and entrapment efficiency of microspheres were used for evaluation indexes.Results The optimum preparation process was the amount of chitosan with 2%,mass ratio of oxymatrine and chitosan with 1∶1,amount of glutaraldehyde with 1 mL and emulsion cross-linking time with 2 h.The drug loading and entrapment efficiency of microspheres were12.56% and 41.98%.Conclusion The preparation of oxymatrine sustained-release microspheres optimizated by orthogonal design could be a simple,quick and accurate method.
