摘    要:目的:探讨塞来昔布对喉癌细胞株Hep-2生长抑制及对环氧化酶-2(COX-2)和Caspase-3变化的影响。方法:将不同浓度塞来昔布作用于体外培养的人喉癌细胞株Hep-2,MTT法计算细胞生长抑制率,流式细胞术检测细胞周期变化,Western blot检测COX-2表达,分光光度法检测Caspase-3活性。结果:塞来昔布呈剂量依赖性显著抑制Hep-2细胞生长,阻滞细胞G0/G1期转化,且可明显降低COX-2蛋白水平,增加Caspase-3活性。结论:塞来昔布能抑制喉癌细胞的生长,其机制可能与下调COX-2表达、增强Caspase-3活性有关。

Abstract:Objective To investigate the effects of celecoxib on the growth of laryngocarcinoma Hep-2 cell line,the expression of COX-2 and Caspase-3 activity.Methods The laryngocarcinoma Hep-2 cell lines were cultured and treated with different concentrations of celecoxib in vitro.The growth inhibitory rate was caculated with MTT method,the cell cycle change was observed with flow cytometer,the expression of COX-2 was detected by Western blot,the activity of Caspase-3 was determined with spectrophotometric method.Results The growth of Hep-2 was significantly inhibited by celecoxib with a dose depedent manner,the cell cycle translation from G0 to G1 was blocked,the expression of COX-2 was significantly decreased,the activity of Caspase-3 was significantly increased.Conclusion Celecoxib could inhibit the growth of laryngocarcinoma cells,which may relate to its down-regulation of COX-2 expression and enhancement of Caspase-3 activity.
