基原对柴胡急性毒性和皂苷类物质含量影响的实验研究

摘    要:目的对不同基原柴胡的皂苷类物质含量和急性毒性大小进行比较研究,探讨基原对柴胡皂苷类化学成分含量与急性毒性的影响。方法运用UV法、HPLC法、经典急毒实验法,进行南、北柴胡药材中柴胡总皂苷和皂苷a含量测定及急性毒性比较研究。结果北柴胡药材中总皂苷含量4.61%,柴胡皂苷a的含量0.60%,南柴胡药材中总皂苷含量3.53%,柴胡皂苷a的含量0.46%。1日内给小鼠灌胃南、北柴胡药材分别为25.2g/kg、26.4g/kg(按含生药量计算),分别相当于临床70kg的成人每公斤体重日用量的196倍和205倍以上,观察14天,小鼠一般状况良好,无一死亡。结论北柴胡药材毒性大于南柴胡药材,其毒性的大小与柴胡皂苷类物质含量有一定相关性。

Abstract:Objective By compare the saponins substances contents with acute toxicity of different original, to approach the effect of the saikosaponin contents and acute toxicity caused by different original. Methods UV methord, HPLC methord and the classical method of acute toxicity ,were used to determine the total saikosaponins contents and the saikosaponin a contents and investigate the acute toxicity of Radix Bupleuri and Radix Bupleuri Scorzoneraefolii. Results The total saponins contents of Radix Bupieuri was 4.61%,the saikosaponin a contents of Radix Bupieuri was 0.60%, the total saikosaponins contents of Radix Bupleuri Scorzoneraefolii was 3.53%,the saikosaponin a contents of Radix Bupleuri Scorzoneraefolii was 0.46%,give the Radix Bupleuri Scorzoneraefolii and Radix Bupieuri to mouse by intragastric administration every day, the dosage is 25.2g/kg and 26.4g/kg discern, which is equal to more than 196 and 205 times about one clinical adult discern, observed 14 days,the general state was well and no one died. Conclusion The toxicity of Radix Bupieuri was greater than Radix Bupleuri Scorzoneraefofii, there is invariably dependability about toxicity and the saikosaponins contents.
