黄芩甙对内皮细胞保护作用及其生化机理

摘    要:为探讨中药黄芩抗细菌感染及其毒血症作用的可能机制,本实验以培养内皮细胞为实验对象,以细菌内毒素为致伤剂,以中药黄芩的有效成分之一一黄芩甙为保护剂,对内毒素损伤内皮细胞以及黄芩甙可能的保护作用,从形态学和生物化学角度进行了探索.结果显示,内毒素主要引起包括线粒体肿胀、内质网扩张、细胞膜崩解在内的膜性结构的损伤,以及细胞连接破坏;黄芩甙能减轻内毒素对内皮细胞膜结构的损伤作用,尤其对细胞连接的保护作用较为明显.同时发现黄芩甙能抑制内毒素所致的细胞蛋白分泌功能增强,蛋白水解酶的释放以及氧自由基的产生等,故认为黄芩甙在一定程度上对内毒素作用的内皮细胞有保护作用.其机理与黄芩甙抑制了内毒素引起的内皮细胞的呼吸暴发和氧自由基的释放等机制有关.

Abstract:In order to study the possible mechanism of Radix Scutellari-ae in antagonising bacterial infection and toxaemia,the cultured bovine pulmonary endothelial cells (BPEC),Endotoxin(ET) and Baicalin were used as orderly the observing object, the injuring and protective factors in this study. The morphologic and biochemical changes of BPEC incubated with ET or Baicalin were observed. The results show that the ET can induce the injury of structure of BPEC including the swelling of mitochodria,thc expanding of en-doplasmic reticulum,collapsing of cell membrane and destroying of the junction between BPEC (Baicalin can reduce the destruction of membrane, especially of the junction of BPEC induced by ET. Baicalin can also inhibit the secretion of protein,the release of pro-teinase and production of free radical of BPEC preincubated with ET. It suggests that Baicalin can partially protect BPEC from injury of ET. Its mechanism is related to the mechanism that the baicalin can inhibit the respiretory burst and release of free radical of BPEC induced by ET.
