湖北汉族人群 PPARγ 基因多态性与妊娠期糖尿病的相关性研究

摘    要:目的探讨PPARγ基因Pr012A1a多态性与妊娠期糖尿病(GDM)的相关性。方法选择156例GDM患者,平均年龄30.72岁:180例正常对照孕妇.平均年龄28.91岁。应用聚合酶链反应-限制性内切酶(PCR-RFLP)技术.对GDM和正常对照孕妇PPARγ基因Pro12Ala多态性位点进行基因分型,同时应用全自动生化分析仪测定血总胆固醇(TC)、甘油三酯(TG)、空腹血糖(FBG)。放射免疫法测定空腹胰岛素(FINS)水平,并计算胰岛素抵抗指数(HOMA-IR)。结果PPARl基因Pr012A1a多态性基因型频率PP:PA:AA在GDM组和正常孕妇组分别为92.4%:7.6%:0.0%和85.0%:13.3%:1.7%.P等位基因和A等位基因在GDM组和正常孕妇组频率分布分别为96.2%、91.7%和3.8%、8-3%。两组间的基因型和等位基因频率差异均无统计学意义(P〉0.05),但PP基因型孕妇的体质量指数明显高于PA基因型(P〈0.01)。结论PPARγ基因Pr012A1a多态性可能与湖北汉族人群GDM的发生无相关性.但与GDM妇女肥胖的发生有一定联系.

Abstract:Objective To study the correlation of PPARγ gene Prol2Ala polymorphism with gestational diabetes mellitus(GDM). Methods A total of 156 patients(mean age of 30.72 years old) with GDM(GDM group) and 180 cases of normal pregnant women (mean age of 28.91 years old, normal control group) were enrolled in the present study. The PPARγ gene Pro12Ala polymorphisms of GDM and normal controls were genotyped by polymerase chain reaction restriction(PCR-RFLP) technology. At the same time, blood total cholesterol(TC), triglyceride(TG), fasting blood glucose(FBG) were measured by automatic biochemical analyzer, fasting insulin (FINS) levels was detected by radioimmunoassay, and insulin resistance index(HOMA-IR) was counted. Results The PPARγ Prol2Ala polymorphism genotype frequencies ofPP : PA : AA in GDM group and normal control group were 92.4 % : 7.6 % : 0 % and 85.0 % : 13.3 % : 1.7 %, respectively. The P allele and A allele in the frequency distribution of GDM group and normal control group were 96.2 %, 91.7 % and 3.8 %, 8.3 %, respectively. Between the 2 groups, the genotypes and allele frequency differences were not statistically significant (P 〉 0.05), but the body mass index of PP genotype was significantly higher than that of PA genotype in pregnant women(P 〈 0.01). Conclusion It is demonstrated that the PPARγ gene Prol2Ala polymorphism may not be related to the incidence of GDM in Han population in Hubei, but obese GDM women have some certain connection.
