Abstract:Objective To establish a HPLC method to the assay of protocatechuie acid in Zhidai granules. Methods Protocatechuic acid was determined by Hypersil Cls column (4.6 mm ×250 mm, 5 μm) using methanol-water-acetic acid( 19:80:1 ) (~)as mobile phase with the flow rate of 1.0 mL ·min^-1 and the detection wavelength was 260 nm. Results There was a good linear relationship between the concentration and peak area value of protocatechuic acid in the range of 0. 051 2 -0. 768 0 μg (r = 0. 999 8). The average recovery was 100. 90% with the RSD of 1.79% ( n = 6 ). Conclusion This method is specific, reliable, reproducible and feasible, which can be used for quality control of Zhidai granules.
