摘    要:目的 观察凋亡蛋白酶活化因子-1基因(Apaf-1)及结肠腺瘤样息肉病易感基因(APC)启动子甲基化状态在膀胱癌组织中的表达,并分析其表达变化与临床资料之间的关系.方法 建立SYBR Green Ⅰ实时定量聚合酶链反应(PCR)的检测方法,通过标准曲线对Apaf-1及APC在36例膀胱癌和15正常组织中的启动子甲基化状态进行定量检测,分析其启动子甲基化量与患者临床病理特征的关系.结果 Apaf-1在膀胱癌组织和正常组织中甲基化率分别为100.00%(36/36)和6.77%(1/15),差异有统计学意义(P<0.01).APC在膀胱癌组织和正常组织中甲基化率分别为69.44%(25/36)和33.33%(5/15),差异有统计学意义(P<0.05).Apaf-1及APC的甲基化量与肿瘤分化程度及临床分期有明显的相关性,随肿瘤分级、分期的增加逐渐升高.结论 Apaf-1及APC的启动子甲基化是膀胱肿瘤发生的早期事件,且与肿瘤的进展有关.

The promoter methylation status of the suppressor genes Apaf-1 and APC in bladder carcinoma tissues
Abstract:Objective To observe the promoter methylation status of the genes apoptotic protease activating factor-1 (Apaf-1) and adenomatous polyposis coli (APC) in bladder carcinoma and analyze the relationship between the expression and the clinical data. Methods The method of SYBR Green Ⅰ quan-titative real-time PCR was established.The expression of tumor suppressor genes Apaf-1 and APC in 36 bladder carcinoma tissues and 15 normal bladder tissues were quantitated by the method of standard curve. The relationship between methylation status of the two suppressor genes and the clinical data were analyzed statistically. Results The methylation rate of Apaf-1 in bladder carcinoma tissues and normal bladder tis-sues were 100.00% (36/36) and 6.77% ( 1/15 ) respectively with statistically significant difference (P < 0. 01). The methylation rate of APC in bladder carcinoma tissues and normal bladder tissues were 69.44% (25/36) and 33.33% (5/15) respectively,with statistically significant difference (P <0.05). The methylation levels were correlated well with the different stages and grades of the tumor and increased with the development of tumor grade and stage significantly. Conclusion The methylation of tumor sup-pressor genes Apaf-1 and APC may have an important role on the early stage of bladder carcinoma, and may also involve in tumor progression.
