化痰散结中药对甲状腺细胞增殖与凋亡影响的实验研究

摘    要:目的:研究化痰散结中药甲肿消对甲状腺FRTL细胞增殖与凋亡的影响.方法:用水煮浸渍提取方法制备甲肿消提取液,甲状腺FRTL细胞株采用含6H的F12培养基培养.中药提取液作用培养细胞,以MTT法测定不同浓度甲肿消对FRTL细胞的增殖影响;采用Annexin V/PI双染法,应用流式细胞仪检测细胞凋亡;P1单染,流式DNA定量法检测细胞周期.结果:甲肿消作用24 h,细胞半数抑制浓度IC50=0.8 mg/ml(含生药浓度),最大抑制率浓度为4.0 mg/ml.甲肿消作用后G0/G1期细胞增多,S和G2/M期细胞减少,增值指数(PI)显著降低,细胞凋亡增加;同时发现甲肿消对细胞增殖与凋亡的影响与作用剂量、作用时间成正相关.结论:甲肿消通过阻滞细胞从G0/G1期向S期转换,从而抑制细胞增殖,同时可诱导FRTL细胞的凋亡,并与作用剂量和作用时间正相关.
