摘    要:目的建立一套快速有效的测量NO的方法并用来监测降压药硝普钠在小鼠体内释放NO的过程。方法建立并采用鲁米诺一过氧化氢(Luminol-H2O2)化学发光体系,动态监测小鼠静脉注射硝普钠(2mg/kg)后血样中NO浓度的变化。结果Luminol-H2O2化学发光体系构建成功,以此测量NO标准系列线性良好;硝普钠给药后,小鼠体内NO30s左右达到峰值,200s恢复给药前水平。结论Luminol-H2O2化学发光体系测量NO灵敏度高、特异性好;硝普钠在体内快速释放NO,硝普钠舒张血管降低血压的作用是通过释放NO实现的。

Abstract:Objective To establish an effective system to detect the nitric oxide release procedure in the rats after intravenous administration of sodium nitroprusside. Methods A luminol H 2O 2 chemiluminescence system was established to continuously detect the changes of blood nitric oxide (NO) in the rats after intravenous injection of sodium nitroprusside (2 mg/kg). Results The luminol H 2O 2 chemiluminescence system was established successfully with a good linearity to NO standard series. After intravenous injection of sodium nitroprusside, the NO in the rats reached its peak at 30 s and returned to the level at 200 s before administration. Conclusion The luminol H 2O 2 chemiluminescence system has a high sensitivity and good specificity to NO, and the results indicate that sodium nitroprusside takes effects by releasing NO in the body.
