低分子肝素对肝癌细胞株HepG_2迁移率及黏附率的影响

摘    要:目的探讨低分子肝素(LMWH)对肝癌细胞株HepG2转移及黏附的影响。方法不同浓度LMWH作用于HepG2细胞株,分别在作用24、48、72 h采用四甲基偶氮唑蓝(MTT)法检测其细胞生长抑制率,Transwell小室检测HepG2细胞迁移率的变化,纤维黏附蛋白黏附实验观察HepG2细胞株的黏附性变化。结果 0.0×106IU·L-1组24、48、72 h的细胞生长抑制率比较差异无统计学意义(P>0.05);0.5×106、1.0×106、1.5×106、2.0×106、2.5×106IU·L-1组24、48、72 h的细胞生长抑制率两两比较,差异均有统计学意义(P<0.05)。同一时间段各浓度组与0.0×106IU·L-1组比较细胞生长抑制率差异均有统计学意义(P<0.05);2.0×106IU·L-1和2.5×106IU·L-1组与0.5×106IU·L-1组比较细胞生长抑制率差异有统计学意义(P<0.05),其余各浓度组两两比较差异均无统计学意义(P>0.05)。与空白对照组比较,0.5、5.0、10.0 g·L-1组细胞迁移率和黏附率下降,差异均有统计学意义(P<0.05);0.5、5.0、10.0 g·L-1组之间细胞迁移率和黏附率两两比较,差异均有统计学意义(P<0.05)。结论 LMWH可抑制HepG2细胞生长并降低其迁移率和黏附率。

Effect of low molecular weight heparin on the migration and adherence of hepatocellular carcinoma cell lines HepG2

Abstract:Objective To explore the effect of low molecular weight heparin( LMWH) on adhesion and migration of hepatocellular carcinoma cell lines HepG2. Methods The different concentration LMWH were added in HepG2cells;then the migration and adhesion of HepG2 cells were examined by Transwell and fibronectin protein test respectively at 24,48 and 72 hours after the LMWH were added in HepG2 cells. Results There was no statistic difference of cell growth inhibition rate in0. 0 × 10^6IU·L^-1 group among 24,48 and 72 hours(P〈0. 05);there were statistic differences of cell growth inhibition rates in 0. 5 × 10^6,1.0 ×10^6,1.5 ×10^6,2.0 ×10^6,2.5 ×10^6IU·L^-1group among 24,48 and 72 hours(P〈0. 05). There were statistic differences of cell growth inhibition rates between 0. 0 × 10^6IU·L^-1 group and 0. 5 × 10^6,1. 0 ×10^6,1. 5 ×10^6,2. 0 × 10^6,2.5 ×10^6IU·L^-1 group(P〈0. 05);there were statistic differences of cell growth inhibition rates between 2. 0,2. 5 × 10^6IU·L^-1 group and 0. 5 × 10^6IU·L^-1 group(P〈0. 05);there was no statistic difference of cell growth inhibition rate among other concenthation groups(P〉0. 05). Compared with the blank control group,the migration and adherence rates in 0. 5,5. 0 and 10. 0 g·L^-1 group were decreased significantly( P〈0. 05);there were statistic differences of the migration and adherence rates among 0. 5,5. 0 and 10. 0 g·L^-1 group(P〈0. 05). Conclusion LMWH can inhibit the growth of cells,and reduce the migration and adherence rates of HepG2 cells.
