摘    要:目的探讨落新妇苷对大鼠肝脏缺血再灌注损伤的保护作用。方法 SD大鼠,分为Sham组(假手术组)、HIRI组(缺血再灌注组)、落新妇苷(低剂量组、中剂量组、高剂量组),建立大鼠肝脏缺血再灌注模型,分别于再灌注4 h、8 h、16 h后取血液及肝脏标本。检测血清谷丙转氨酶(ALT)、谷草转氨酶(AST);光镜下观察肝细胞显微结构变化;免疫印迹试验(Western Blot)分析肝组织中IL-1、caspase-3、-8、-9的表达;流式细胞仪检测肝细胞凋亡率。应用SPSS l5.0统计软件进行分析,计量资料以均数±标准差(±s)表示,对各组大鼠血清ALT、AST,肝组织中IL-1、caspase-3、-8、-9蛋白的表达及细胞凋亡情况进行比较采用单因素方差分析和t检验,以P0.05、P0.01为差异有统计学意义。结果 HIRI组ALT和AST水平升高,而药物组明显降低。光镜结果显示药物组肝细胞损伤明显减轻。Western Blot结果显示,高剂量组IL-1、caspase-3、-8、-9蛋白的表达较HIRI组明显减轻。高剂量组的细胞凋亡率也较HIRI组降低。结论落新妇苷预处理可减轻大鼠肝脏缺血再灌注损伤,其作用机制可能与其抑制caspase-3、-8、-9的表达,从而抑制细胞凋亡有关。

Astilbin alleviates hepatic ischemia-reperfusion injury in rats by downregulating the expression of caspase-3, -8, -9
Abstract:Objective To explore the protective effect of astilbin in hepatic ischemia-reperfusion injury(HIRI). Methods SD rats are randomly divided into Sham group (control group), HIRI group ( ischemia-reperfusion group ) , astilbe ( low dose group , middle dose group , high dose group ) , and model of rat HIRI was established .Liver specimens and blood of rats were harvested at 4 h, 8 h, 16 h after reperfusion , in order todetect serum alanine aminotransferase ( ALT ) , aspertate aminotransferase ( AST ) , change of liver cell microstructure , the expression of IL-1, caspase-3、-8、-9 in liver tissue by Western Blotting and liver cell apoptosis rate by Flow cytometry .Statistical analysis were perfromed by using SPSS 15.0 software.Measurement data were expressed as (x±s), and were examined by single factor analysis of variance and t test, including serum ALT, AST、the expression of liver tissue IL-1, caspase-3、-8、-9 and apoptosis rate.When P<0.05, P<0.01 difference were considered to be statistically significant . Results The level of ALT , AST in HIRI group were increased significantly , while those in drug groups were decrease .The results of Light microscope indicate that liver cell damage in drug groups were significantly decreased.The results of Western Blot showed that expression of IL-1, caspase-3、-8、-9 were significantly reduced in high dose group , compared with HIRI group .Apoptosis rate in High dose group were also decreased . Conclusion Astilbe pretreatment could reduce HIRI and its mechanism might be associated with inhibiting the expression of caspase-3、-8、-9, and decreased apoptosis of liver cells .
