摘    要:目的探讨低浓度H2O2对同步化处理或未经处理的HL-60细胞作用的时间效应关系. 方法将培养的HL-60细胞进行同步化处理或取对数生长期细胞,施加不同浓度梯度的H2O2影响,作细胞计数,四唑盐(MTT)比色试验检测细胞生存率,流式细胞技术测定细胞周期分布. 结果低浓度H2O2作用组HL-60活细胞数明显高于未施加影响组,尤以H2O2终浓度为2 μmol*L-1组作用明显(P<0.01),细胞周期也有轻度改变. 结论低浓度H2O2作用HL-60细胞可使细胞数增加,其影响有时间效应关系,机制可能同其影响细胞周期有关.

Abstract:AIM To investigate the effects of hydrogen peroxide on human leukemic cells (HL-60) in vitro, and illustrate its possible mechanism. METHODS Cultivated HL-60 cells of synchronization or non-synchronization were affected by different concentration of hydrogen peroxide. Then the number of living cells was recorded and the vitality of HL-60 cells was measured by MTT methods. Cell cycle distribution was analyzed by DNA flow cytometry. RESULTS Compared with the control group, the number of HL-60 cells affected by low concentration of hydrogen peroxide was increased. The effect on the Group of 2 μmol*L-1 was especially remarkable. The cell cycle changed slightly. CONCLUSION Low concentration of hydrogen peroxide could increase the number of HL-60 living cells. Its main mechanism might probably be involved in the change of the cell cycle.
