吴茱萸碱对大鼠肝癌细胞缝隙连接细胞通讯功能及连接蛋白表达的影响

摘    要:目的探讨吴莱萸碱抗肿瘤的作用机制。方法用终浓度为0.1,0.2,0.3μmol/L的吴茱萸碱作用于CBRH7919细胞,同时设不加吴茱萸碱的空白对照组。采用划痕标记染料示踪技术检测各组细胞缝隙连接细胞通讯(gap junctional intercellular communication,GJIC)功能;异硫氰酸荧光素间接免疫荧光法检测连接蛋白(connexin,Cx)26、Cx32表达的变化。结果与空白对照组相比,吴莱萸碱各浓度组CBRH7919细胞Cx26、Cx32表达显著增加,GJIC功能增强,并呈一定的剂量效应依赖关系。结论吴茱萸碱上调CBRH7919细胞Cx26和Cx32的表达及促进CBRH7919细胞的GJIC功能,可能是其抗肿瘤的机制之一。

Abstract:Objective To explore the mechanism of evodiamine in treating Carcinoma.Methods The rat hepatocarcinoma cells CBRH7919 were treated by evodiamine with final concentration at 0.1,0.2 and 0.3 μmol/L for 48 hours.Simultaneously,evodiamine untreated CBRH7919 cells were used as blank control.The function of GJIC in each group was detected by scrape-loading dye transfer assay.The expression of connexin(Cx) 26 and Cx32 in CBRH7919 cells of each group was detected by fluorescein isothiocyanate labeling indirect immunofluorescence.Results Compared with those in blank control group,the expression of Cx26 and Cx32 in CBRH7919 cells in all groups treated with different concentration of evodiamine was significantly increased,and the function of GJIC was significantly enhanced with a dose dependent manner.Conclusion One of antineoplastic mechanisms of evodiamine maybe associate with its up-regulating the expression of Cx26 and Cx32 and promoting the function of GJIC in CBRH7919 cells.
