胡椒碱对过氧化氢损伤大鼠心肌线粒体的保护作用及机制研究

摘    要:目的研究胡椒碱抗心肌抗缺血、增强耐缺氧能力及心肌保护作用的机制。方法采用过氧化氢损伤大鼠心肌线粒体模型,观察胡椒碱对心肌线粒体中谷胱甘肽(GSH)、丙二醛(MDA)、ATP酶和门冬氨基转移酶(AST)的影响。结果过氧化氢损伤后,大鼠心肌线粒体中MDA水平升高,GSH含量降低,Na^+K^+-ATP酶和Ca^2+-ATP酶活性降低,AST活性升高,而胡椒碱可以抑制MDA生成、增加AST活性,提高GSH含量并增强Na^+K^+-ATP酶和Ca^2+-ATP酶活性。结论胡椒碱可能是通过升高心肌线粒体GSH,降低MDA及增加Na^+K^+-ATP酶和Ca^2+-ATP酶活性来实现其心肌保护作用。

Abstract:Aim To investigate the effects and mechanisms of piperine in protecting against myocardial ischemia and enhancing the levels of antioxidants in rats. Methods Myocardial mitoehondrial injury in rats was induced by hydrogen peroxide in vitro. The MDA, GSH, Na ^+K^+ -ATPase, Ca^2+ -ATPase and AST levels were detected. Results Piperine reversed oxidative stress injury of myocardial mitochondrial induced by hydrogen peroxide( P 〈0. 05),prevented the loss of GSH and the increase of MDA,advanced the activity of Na ^+K ^+ -ATPase and Ca^2+ -ATPase ,and maintained the normal activity of AST. Conclusion Piperine may exert myocardial preservation effects by increasing myocardial mitochondrial the antioxidant agents,inhibitting the lipid peroxidation and oxidation enzyme,and increasing the activity of ATPase to maintain the ionic balance between the intra-cellular and extra-cellular in myocardial ischemia.
