三物白散抗肿瘤免疫正相调节机制的实验研究

摘    要:目的:在整体水平与细胞分子水平上研究三物白散抗肿瘤过程中免疫正相调节的作用机制。方法:建立小鼠腋下移植性肿瘤模型,三物白散谩大、中、小3个剂量组,另设一个正常对照组(生理盐水组)和一个阳性药组(5-氟尿嘧啶组),用药8天后,摘取皮下瘤块、胸腺、脾脏,称其重量,计算抑瘤率、脾脏指数、胸腺指数,另摘眼球采血取血清,用放射免疫检测法检测荷瘤小鼠血清中IL-2、IL-4表达;分离BALB/C小鼠的胸腺细胞,经三物白散作用后,检测其IL-2、IL-4表达。结果:与正常对照组相比,用药组的体内抑瘤率均有明显差异(P〈0.01);三物白散各组的胸腺、脾脏重量与正常对照组无明显差异,5-Fu组胸腺、脾脏重量与正常对照组相比差异显著(P〈0.05);与正常对照组相比,体内试验三物白散各剂量组IL-2水平升高(P〈0.01),IL-4水平下降(P〈0.01);体外试验除Ⅳ组的IL-2参数外,与体内试验相似。结论:三物白散在整体水平上具有确切的抗肿瘤免疫正相调节作用,其机静可能是通过逆转Th1/Th2漂移来实现的。

Abstract:Objective:To study the regulation effect of Sanwubai Powder(SP) on immunity with tumors on general and cell levels.Methods:The rat model with transplanted tumor in armpit was set up.The SP groups of high dosage,medium dosage and low dosage, the normal control (saline) and the positive control (5-Fludrouracil).The tumor, thymus and spleen were excised after 8 days,the antineoplastic rate,spleen index,thymus index and IL-2,IL-4 expressed were determined.Conclusion:The difference of antineoplastic rate between the SP group and the normal group was significant(P<0.05).The difference of weight of thymus,spleen between SP group and the normal group was not significant.The difference of weight of thymus,spleen between 5-Fludrouracil group and the normal group was significant(P<0.05).IL-2 level was raised and IL-4 was reduced in SP group compared with the normal group(P<0.01).Conclusion:There was definitely positive effect of SP on immunity with tumor.
