生长抑素对裸鼠肝部分切除后肝癌组织血管内皮生长因子表达及新生血管形成的影响

摘    要:目的 观察生长抑素对肝部分切除裸鼠肝癌组织微血管密度 (MVD)和血管内皮生长因子 (VEGF)表达的影响 ,探讨其对肝部分切除后残肝肿瘤生长的影响。方法  3 0只裸鼠随机均分成 3组 :A组 (对照组 ) ,B组 (肝部分切除组 )和C组 生长抑素(SST)治疗组 ] ,A组仅将人肝癌组织种植于右肝叶 ,B组和C组切除肝左叶和中叶后于右肝叶种植人肝癌组织 ,C组术后第一天开始腹腔注射SST(2 5 0ug/kg ,bid) ,B组和C组腹腔注射等量的生理盐水 ,3组动物于 3 5d后处死 ,检测肿瘤大小和重量 ,采用免疫组化和图像分析法检测MVD和VEGF的表达。结果 B组肝癌组织MVD和VEGF表达较A组明显增强 (P <0 0 5 )肿瘤重量和体积亦明显增加 (P <0 0 5 ) ,C组肝癌组织MVD和VEGF表达较B组和A组明显减低 (P <0 0 5 )肿瘤重量和体积亦明显减少 (P <0 0 5 )。结论 SST可以通过拮抗VEGF的表达 ,抑制肿瘤新生血管形成而减轻肝部分切除促肿瘤生长的作用。

The Effects of Somatostatin on the VEGF Expression and Angiogenesis of Liver Cancer Tissues after Partial Hepatectomy in Nude Mice

Abstract:Objective To observe the effect of somatostatin(SST) on the VEGF expression and MVD of tumor tissues in nude mice after partial hepatectomy . Methods 30 nude mice were randomly divided into three groups:groupA(control group), group B ( partial hepatectomy group), group C (somatostatin treatment group). In group A, human hepatocellular carcinoma(HCC) tissue was implanted into right liver lobe, and in group B and C, human HCC tissue was implanted into right liver lobe after resection of left and medial liver lobes. In group C,nude mice received SST (250ug/kg.d, bid )intraperitoneal injection from the first day after operation, and in groups A and B,nude mice received the same amount of saline solution intraperitoneal injection.All the mice were killed at the 35th day after operation, the weight and volume of tumors were measured, and MVD and VEGF expression were determined by immunohistochemical staining and image analysis system. Results In group B, MVD and VEGF expression of tumor tissues were markedly higher than those in group A(P<0.05 ),and the weight and volume of tumor tissues were also obviously higher than those in group A (P<0.05). In group C, MVD and VEGF expression were significantly lower than those in groups B and A (P<0.05), and the weight and volume of tumor tissues were also markedly lower than those in groups B and A (P<0.05). Conclusion Somatostatin could inhibit the growth of tumor cells after partial hepatectomy by decreasing the expression of MVD and VEGF and inhibiting tumor angiogenesis.
