钩吻素子对小鼠佐剂性关节炎的治疗作用

摘    要:目的:观察钩吻素子对小鼠佐剂性关节炎的治疗作用。方法筛选成功建立佐剂性关节炎模型的小鼠,随机分为模型组、钩吻素子高(20mg/kg)、中(4mg/kg)、低(0.8mg/kg)剂量及阳性对照药吲哚美辛处理组(10mg/kg),每组12只,另设置12只未免疫正常小鼠为对照组。建模后第8天起,以灌胃方式给药,每天1次,连续10d。以小鼠体质量、注射足厚度、热辐射痛阈热缩足反射潜伏期(PWTL)以及组织病理学变化等为指标,观察钩吻素子的治疗作用。结果小鼠建模后出现显著的体质量减轻、注射足厚度增加、热辐射痛阈下降,苏木精‐伊红(H‐E)染色显示关节结构破坏、滑膜增生以及炎症细胞浸润。钩吻素子灌胃后,小鼠体质量增加,显著高于模型组,注射足厚度显著小于模型组,PWTL显著高于模型组,作用呈剂量依赖性。H‐E染色显示钩吻素子处理组小鼠踝关节的炎性细胞浸润及滑膜增生、骨质破坏等得到改善。结论钩吻素子对小鼠的佐剂性关节炎具有治疗作用。

Abstract:Objective To examine the therapeutic effect of koumine on adjuvant arthritis in mice . Methods Eight days after immunization ,mice with adjuvant arthritis received koumine (0 .8 ,4 ,or 20 mg/kg) ,or indomethacin (10 mg/kg) as control through gastric gavage for 10 consecutive days . Body weight ,injected paw thickness and paw withdrawal thermal latency (PWTL) were examined prior to the immunization and after the initiation of drug treatments on alternate days and followed by histological ex‐amination . Results After immunization ,the body weight decreased ,the injected paw thickness in‐creased and the PWTL decreased in mice with adjuvant arthritis . Histological examinations showed that the immunized mice exhibited apparently joint structure destruction ,synovial hyperplasia and inflammato‐ry cell infiltration . Repeated intragastric administrations of koumine significantly attenuated the above physical and hyperalgesia changes observed in arthritic animals in a dose‐dependent manner and inhibited the histopathological lesion . Conclusion Koumine can effectively attenuate adjuvant arthritis in mice .
