叩击式骨应力刺激仪预防废用性骨质疏松的实验研究

摘    要:目的通过建立一种新的废用性骨质疏松山羊模型,观察叩击式骨应力刺激仪预防废用性骨质疏松的效果。方法4月龄山羊18只,随机分组:①对照组,②废用组,③治疗组。废用组和治疗组切断右侧坐骨神经。治疗组术后运用叩击式骨应力刺激仪进行治疗,叩击力为100N,频率为3Hz。每天3次,每次60min,每次间隔3h。治疗2~4个月后分别检测和分析山羊右侧胫骨骨密度、组织学及生物力学的变化。结果同对照组和治疗组相比,废用组胫骨的骨密度值及生物力学性能明显下降(P0.01);而治疗组与对照组也有差别(P0.05)。表现为:对照组治疗组废用组。组织病理学检查提示:HE染色见废用组骨小梁较对照组和治疗组明显减少、变细。Masson染色结果表明废用组骨基质矿化不良,骨基质中矿物成分较对照组和治疗组明显减少。结论切除山羊一侧坐骨神经可以建立起废用性骨质疏松模型。叩击式骨应力刺激仪对废用性骨质疏松有明显的防治效果。

Abstract:Objective By establishing one new kind of goat disuse osteoporosis model,the preventive effect of the stimulator of bone stress was researched.Methods Thirty 4-month-old goats,were used in this study.Right hindlimbs of 12 goats were performed sciatic neurectomy to become cripple and served as unloading group and treatment group;The 6 goats were served as control group.After operation,the goats in treatment group was received the stress stimulation treatment 3 times every day,60 min every time,the interval time is 3h.After 2-4 months treatment,BMD (bone mineral density),Histology and biomechanics of the right tibials of goats were measured and analyzed.Results Compared with the control group and the treatment group,the tibial BMD and biomechanic nature of the unloading group decreased significantly (P < 0.01).There were the differences between the control group and the treatment group (P<0.05).It was showed:control group>treatment group>unloading group.Histological examination:HE staining shown that the tibial trabecula of the unloading group became less and thinner which was more and thicker in the control group and the treatment group.Masson staining indicated that the unloading group had less bone mineral components than the control group and the treatment group,and its bone matrix had been mll mineralized.Conclusion That the disuse osteoporosis can be prevented effectively by the stimulator of bone stress was researched.
