颗粒细胞在玻璃化冷冻中对卵母细胞的影响

摘    要:目的 采用开放式拉细麦管(open pulled straws,OPS)法玻璃化冷冻技术,探讨卵丘颗粒细胞对未成熟卵母细胞冻融后存活率及其发育能力的影响。方法 小鼠生殖泡期卵丘-卵母细胞复合物(COC)或完全去颗粒细胞后的裸卵(DO)用OPS法玻璃化冻存,复苏后行体外成熟培养(IVM);COC直接行IVM作为对照组。结果 COC及裸卵冻融后存活率(分别为69.49%、77.90%)、成熟率(分别为56.10%、56.74%)无显著性差异,但成熟率均显著低于对照组(82.28%)。结论 在未成熟卵的玻璃化冷冻中颗粒细胞对卵母细胞冻存无保护作用。

Influence of Granulosa Cells on Oocytes during Vitrification
Abstract:Objective To study the influence of granulosa cells on the survival rate and maturation rate of immature oocytes after vitrification by open pulled straws.Methods The cumulus-oocyte complexes or denuded oocytes (DO) at germinal vesicle stage were collected and vitrified,followed by in vitro maturation after thawing. The fresh COC directly subject to in vitro maturation served as control group.Results There was no significant difference in the survival rate and maturation rate between COC and DO after vitrification (69.49% vs 77.90%; 56.10% vs 56.74% respectively). The maturation rate in treatment groups was significantly lower than in the control group (82.28%,P<0.05). Conclusion Granulose cells have no protective effects to oocytes during vitrification.
