舌鳞癌角蛋白单抗的制备与鉴定

摘    要:目的:以人舌鳞状细胞癌角蛋白组群为抗原,制备了抗舌鳞癌角蛋白单克隆抗体,并对单抗的特异性进行鉴定和免疫组织定位。方法:提取和纯化的舌鳞癌角蛋白免疫小鼠,建立了五株稳定分泌抗体的杂交瘤细胞株。免疫组化方法对18例口腔鳞癌病例进行抗体的特异性鉴定。结果:共获得5株抗角蛋白单克隆抗体杂交瘤细胞株,分别命名为Lck1-5,免疫鉴定Lck1、Lck3、Lck4、Lck5都可在癌组织及癌转移淋巴结中部分表达,表达阳性细胞位于癌巢中的癌细胞胞浆中。其中以Lck4的特异性和敏感性最高。结论:成功进行了舌鳞癌角蛋白单克隆抗体的制备,其中Lck4敏感度、特异度最高。

Abstract:Objective:To use tongue squamous cell carcinoma keratins as antigen to prepare the monoclonal antibody,and to evaluate the immunohistochemical location with the monoclonal antibody.Methods: Immunize BALB/c mice by B leukomonocyte hybridoma technique,five cloned strain had been acquired.18 cases of oral squamous cell carcinoma specimens were pathologically chosen and stained by S-P microwave immunohistochemical method.Results: Those five hybridoma cell strains which could secrete keratin monoclonal antibody of anti-tongue squamous cell carcinoma stably were named from Lck1 to Lck5.Lck1,Lck3,Lck4 and Lck5 could express partly in carcinoma tissue and metastasis lymphonode.Positive cells were located in cancer cells cytoplasm.Specificity and sensitivity of Lck4 were the highest.Conclusions: 5 strains of monoclonal antibody are acquired.Specificity and sensitivity of Lck4 are the highest in 5 strains of monoclonal antibody.
