摘    要:目的 探讨缺血性脑水肿后水通道蛋白4(AQP4)的变化,研究脑水肿的发病机制。方法 选用Wis—tar雄性大鼠42只,体重250~300g,鼠龄3~4月,按缺血时间随机分为6h、1d、3d、5d、7d、14d及假手术组,参考Lon—ga等方法建立大鼠大脑缺血模型,应用免疫组化染色检测脑组织中AQP4的变化。结果 AQP4在缺血组织表达增强,缺血6h即增强,3d达高峰,7d恢复正常。结论 AQP4是影响缺血性脑水肿的重要因素。

Abstract:Objective To explore the change of AQP4 in post ischemic edema and to research the pathogeny of intracranial edema. Methods 42 healthy male Wistar rats were divided randomly into shamed operation subgroup and 6h,1d,3d,5d,7d and 14d subgroups according to the time exposed to ischemia. The model of middle cerebral artery occlusion was performed with intraluminal filament occlusion. The sample were analyzed by immunohistochemical methods. Results Expression of AQP4 increased after MCA occlusion,and increased significantly on 3d,and get right on 7d.Conclusion AQP4 plays a role in post ischemic edema formation.
