摘    要:目的:观察三氧化二砷(As2O3)对NB4细胞珠端粒酶活性的影响,并探讨与细胞增殖、分化及细胞周期之间的关系。方法:应用MTT法、BNT法检测细胞的增殖抑制率、细胞分化率、流式细胞仪检测CD11b的表达率、细胞周期时相比率,以PCR为基础的TRAP法检测端粒酶活性。结果:As2O3能有效地使NB4细胞增殖抑制、诱导分化,伴随着端粒酶活性的变化为活性降低。细胞周期分析发现,G0/G1期高或S期低对应低水平端粒酶活性,G0/G1期低或S期高对应高水平端粒酶活性。结论:As2O3可抑制NB4细胞端粒酶活性,并与细胞分化、细胞周期时相有关。

Abstract:Objective:To study the telomerase activity in NB4cells exposed to arsenic trioxide(As 2 O 3 ).Methods :The inhibition of cell proliferation,induction of cell differentiation,CD11b expression,mitotic cycle and telomerase activity were evaluated in NB4cells exposed to As 2 O 3 .Results:Telomerase activity was remarkably inhibited in NB4cells exposed to As 2 O 3 accompanied by inhibition of cell prolifera-tion,induction of cell differentiation and CD11b expression.A long G0/G1phase or a short S phase coincided with low telomerase activity while a short G0/G1phase or a long S phase coincided with high telomerase activity.Conclusion:As 2 O 3 can lower the telomerase activity in NB4cells,which has a close relationship with induction of cell differentiation and mitotic cycle.
