苯纳普利对大鼠残肾模型肾脏细胞凋亡及Bcl-2、Bax表达的影响

摘    要:目的 探讨血管紧张素转换酶抑制剂 (ACEI)对肾脏病变渐进性发展过程中细胞凋亡及凋亡相关基因表达的影响。方法 SD大鼠 5 /6肾切除后 ,每日灌胃苯纳普利 ( 10mg/kg) 12周 ,采用流式细胞术 ,TUNEL和免疫组织化学技术检测肾脏细胞凋亡及Bcl 2、Bax表达情况。结果 肾切除组细胞凋亡及Bcl 2、Bax表达均高于假手术组 ;苯那普利治疗组细胞凋亡减少 ,Bax表达减弱 ,Bcl 2表达增强。结论 苯纳普利能抑制肾脏病变进展过程中的细胞凋亡 ,影响凋亡相关基因可能是其作用途径之一。

Effects of benazepril on apoptosis and the expression of Bcl-2 and Bax in the remnant kidney model
Abstract:Objective To investigate the effects of angiotensin converting enzyme inhibitor benazepril on apoptosis and the expression of Bcl 2 and Bax in the rat remnant kidney model. Methods Remnant kidney model was induced in adult male SD rats by 5/6 nephrectomy. Benazepril(10 mg/kg) was delivered daily by gavage. Apoptosis was evaluated by means of TUNEL and flow cytometry. The expression of Bcl 2 and Bax was detected by immunohistochemistry and flow cytometry. Results Compared with those in the kidneys of sham operated group, the number of apoptotic cells was more and the expression of Bcl 2 and Bax was higher in the remnant kidneys. Benazepril caused a decrease in apoptotic phenomena and expression of Bax , and an increase in expression of Bcl 2. Conclusion Benazepril can inhibit apoptosis in remnant kidneys and regulating the expression of apoptosis related genes may be one of the mechanisms.
